目的:观察人的唇腺中CCL28的表达和分布,以期探讨CCL28在唇腺iga分泌中的作用。方法:应用实时荧光定量PCR对13例唇腺标本中的CCL28mRNA的表达进行检测,以已知有CCL28表达的大肠组织作为对照;并将CCL28在唇腺与腮腺中的表达进行比较。用免疫组化法对13例唇腺标本中的CCL28蛋白质分布进行定位。结果:人的唇腺中表达高水平的CCl28mRNA,并且唇腺中表达量(1.28±0.57)高于腮腺(0.79±0.24)(P〈0.01)。在免疫组化中,唇腺腺泡上皮细胞、腺管上皮细胞和腺泡内黏液CCL28染色呈阳性,但染色深度不同。结论:唇腺中表达CCL28mRNA。CCL28蛋白由唇腺腺泡上皮细胞产生,并最终分泌到黏液内。结合唇腺在口腔IgA分泌中的重要作用,认为唇腺分泌高浓度IgA可能与CCL28的高表达有关。
AIM: To detect the expression and distribution of CCL28 in human labial glands, and to explore the role of CCL28 in IgA secretiion by labial glands. METHODS: CCL28 mRNA was detected by real-time PCR, while CCL2.8 protein was detected by immunohistochemistry (IHC). Labial gland samples were from 13 healthy adults. Large intestine sample served as PCR positive control . CCL28 expression between labial glands and parotids were compared. RESULTS: CCL28 mRNA was abundantly expressed in labial glands of healthy adults. CCL28 expression ( 1.28 ± 0.57 ) in labial glands was higher than that in parotids (0.79 ± 0.24) ( P 〈 0.01 ). Positive staining of CCL28 was identified in serous epithelial cells, duct epithelial cells and mucus, but with different staining intensities. CONCLUSION: CCL28 mRNA is expressed in labial glands. The protein is synthesized in serous epithelial cells, and secrets into mucus, indicating that the high concentration of IgA in labial glands may be associated with the high expression of CCL28.
