目的:旨在观察离心运动致骨骼肌损伤后丝氨酸蛋白酶(Omi)在骨骼肌组织中的表达变化。方法:8周龄雄性SD大鼠36只,随机分为安静对照(n=6)、离心运动2大组(n=30),离心运动组大鼠在跑台上进行一次性离心运动,分别于运动后即刻(E0组,n=6)、运动后12 h(E12组,n=6)、运动后24 h(E24组,n=6)、运动后48 h(E48组,n=6)和运动后72 h(E72组,n=6)麻醉大鼠,腹主动脉取血,取比目鱼肌待测;使用荧光酶联免疫吸附法检测半胱氨酸天冬氨酸特异性蛋白酶-3(Caspase-3)的活性;Western Blot和Realtime-PCR法分别检测Omi、凋亡抑制蛋白(XIAP)的蛋白表达和基因水平;Co-IP法检测XIAP和Omi的相互结合作用。结果:与对照组相比,离心运动后Caspase-3、Caspase-9活性均明显升高;离心运动后Omi、XIAP的蛋白表达和m RNA水平均明显升高;离心运动能明显增加XIAP和Omi的结合量。结论:离心运动能够激活骨骼肌组织Caspase-9和Caspase-3的活性,诱导Omi和XIAP的表达上调,促进XIAP与Omi相互结合,Omi可能在促使凋亡过程中发挥重要介导作用。
Objective:The aim of the present study is to investigate the change of Omi in skeletal tissue injury after eccentric exercise and the underlyingmechanism. Methods:36 Adult SD rats,8 weeks aged,were randomly divided into two groups:control group and five kinds exercise groups which included 0 hafter exercise,12 h after exercise,24 h after exercise,48 h after exercise and 72 h after exercise. The exercise groups were made to run on amotor-driven forone-off eccentric exercise. The rats were sacrificed in batches at different time points after exercise,then the soleues were saved to measure. Caspase-3and-9 were determined by flurometric immunosorbent enzyme assay method. Protein and m RNA levels of Omi,XIAP were determined by Western blot andReal time PCR,respectively. The combination of XIAP and Omi was determined by Co-IP. Results:Compared with those in control group,Caspase-3and-9 activity were significantly increased after eccentric exercise;Protein expression and m RNA levels of Omi,XIAP were significantly increased after ec-centric exercise. Eccentric exercise improved the combination of XIAP and Omi. Conclusions:Eccentric exercise can increase the expression of Omi andXIAP,improve the combination of XIAP and Omi,induce enhanced Caspase-9 and-3 activity and then induce skeletal muscle cell apoptosis.
