目的:研究复方中药岩舒注射液能否抑制人白血病HL-60多药耐药细胞增殖及其相关机制。方法:采用人白血病HL-60敏感细胞及其HR-20耐药细胞进行研究。MTT法检测药物对细胞增殖的影响,流式细胞术检测多柔比星的荧光强度变化,免疫印迹方法检测P-糖蛋白含量的变化。结果:岩舒注射液作用耐药和敏感细胞的Ic50值相近,没有明显的交叉耐药性。岩舒注射液增加高三尖杉酯碱对耐药细胞的增殖抑制作用,增加多柔比星在耐药细胞内的积聚。免疫印迹法检测到:岩舒注射液处理后明显降低耐药细胞的P-糖蛋白水平。结论:岩舒注射液能克服人白血病HL-60细胞的多药耐药性,其机制与降低P-糖蛋白的水平有关。
Objective : To investigate the mechanism underlying the inhibiting effect of Yanshu injection on proliferation of multidrug-resistant human leukemia HL-60 cells. Methods: Human HL-60 and HR-20 resistant cells were treated with Yanshu injection. The inhibition of cell proliferation was detected by MTT assay. The fluo- rescence intensity of doxorubicin was detected by flow cytometry. The level of P-glycoprotein was detected by immu- nblotting. Results: The ICs0 value of Yanshu injection in HR-20 cells was similar to that in HL-60 sensitive cells, indicating no cross resistance. Furthermore, the combination with Yanshu injection obviously increased the inhibito- ry actions of homoharringtonine on cell proliferation. Yanshu injection increased the accumulation of doxorubicin in the HR-20 cells. P-glycoprotein expression was reduced in the HR-20 cells after exposure to Yanshu injection. Conclusion: Yanshu injection can overcome the muhidrug resistance through reducing the level of P-glycoprotein expression in human leukemia HL-60 cells.
