采用电喷雾质谱法(ESI-MS)研究了12-氨基乙酰脱氢枞胺(12-ADA)与5种单链DNA-dA6,dT6,dG6,dC6,d(AT)3的相互作用。考察了12-ADA与这5种DNA以不同的摩尔比(1∶10,1∶1,10∶1)混合时,形成的12-ADA-DNA非共价复合物及其电喷雾质谱行为。结果表明,12-ADA-DNA复合物容易负电离并主要形成-3和-4价离子;12-ADA与各DNA复合物的化学结合计量比随12-ADA和DNA的摩尔比增加而增大。12-ADA与DNA摩尔比从1∶10增大至10∶1时,12-ADA与DNA化学结合计量比从1∶1增大至4∶1。
The interaction between 12-aminodehydroabietylamine acetamide(12-ADA) and 5 different single-stranded DNA(dA6,dT6,dC6,dG6,d(AT)3) was studied by ESI-MS. 12-ADA-DNA complexes and their ESI-MS behavior at different 12-ADA/DNA molar ratio(1∶10,1∶1,10∶1) were also investigated. The results showed that 12-ADA-DNA complexes were easily ionized in ESI negative mode and mainly formed -3,-4 charged ions,and the binding stoichiometries of complex ions were related to the molar ratio of 12-ADA/DNA. The binding stoichiometries of 12-ADA-DNA complex increased from 1∶1 to 4∶1 by increasing the molar ratio of 12-ADA/DNA from 1∶10 to 10∶1.
