目的:建立甲状腺功能低下阳虚模型,为病理研究、药物研发奠定基础。方法:将SD雄性大鼠随机分为正常组、模型组,每天上午定时灌胃薄荷油1.25mL/kg,持续灌胃21d;分别在第14、21天检测血清甲状腺功能、物质能量代谢、肝肾功能等相关指标。结果:薄荷油诱导的阳虚模型组,与正常组相比,在第14天、21天出现三碘甲腺原氨酸(T3)、甲状腺素(T4)显著性降低(P〈0.05,P〈0.01);葡萄糖(GLU)、甘油三酯(TG)、总胆固醇(TC)、白蛋白(ALB)、钠-钾-腺嘌呤核苷三磷酸酶(Na+-K+-ATPase)与正常组比显著性降低(P〈0.05,P〈0.01);而其血清中肌酐(Cr)、尿素氮(BUN)、谷丙转氨酶(ALT)、谷草转氨酶(AST)与正常组比较均没有统计学差异。结论:给大鼠灌胃薄荷油1.25mL/kg,在第14-21天可以出现较理想的甲状腺功能低下的阳虚动物模型,且未引起肝肾毒性反应。
Objective: To establish the hypothyroidism with yang-deficiency model for pathological studies and to lay the foundation for drug research and development.Methods: Male SD rats were randomly divided into normal group and model group,fed regularly every morning,peppermint oil 1.25mL/kg,continuous gavage for 21 days;serum thyroid function,material and energy metabolism,liver and kidney function and other related indicators were tested at day 14 and day 21.Results: Yang-deficiency and peppermint oil-induced model group,compared with normal group,at day 14 day and day 21,developed significant reduction of T3,T4;GLU,TG,TC,ALB,Na+-K+-ATPase,compared with the normal group,were significantly lower(P0.05,P0.01);and there was no significant difference of Cr,BUN,ALT,AST between model group and normal group.Conclusion: The rats,fed with peppermint oil 1.25mL/kg,could become satisfied animal model with hypothyroidism and yangdeficiency syndrome,without inducing liver and kidney toxicity.
