目的观察白藜芦醇对db/db糖尿病鼠视网膜病变的影响。方法选用18周龄db/db糖尿病鼠,db/m型同性同窝瘦型鼠为对照。将db/db鼠随机分为:db/db组,白藜芦醇高(50 mg/kg)、低剂量(10 mg/kg)治疗组,每组6只,药物连续灌胃2个月。观察一般状态,测空腹血糖3次,视网膜电流图(ERG)检测,2个月末处死各组鼠。HE染色观察视网膜病理改变,Western blot检测VEGF蛋白表达。结果与db/db组比较,白藜芦醇治疗组血糖水平显著降低(P〈0.05),尤其以高剂量组为著,HE染色db/db组鼠视网膜内界膜粗糙,厚度变薄,白藜芦醇治疗后显著改善。与db/db组比较,白藜芦醇治疗组主要表现为暗适应3.0反应中a波、b波的潜伏期缩短(P〈0.05)。Western blot结果显示db/db组VEGF蛋白表达显著升高,但白藜芦醇治疗后表达显著降低(P〈0.05)。结论白藜芦醇能降低db/db鼠血糖,改善db/db鼠视网膜功能,其机制可能通过下调血管生成因子VEGF蛋白表达来发挥作用。
Objective To observe the effect of resveratrol on retinopathy of db/db diabetic mice. Methods The 18-week-old db/db diabetic mice and db/m mice( control) were chosen. Db/db mice were randomly divided into three groups( n = 6 in each group) : db/db group,resveratrol high( 50 mg/kg) and low dose( 10 mg/kg) group. Resveratrol were given for 2 months. The states of mice were observed,the fasting blood glucose level was measured three times,and the retinal current( ERG) was detected. Mice were sacrificed at the end of 2 months. HE staining was used to observe the pathological changes of retina and the expression of VEGF protein was detected by Western blot. Results Compared with db/db group,the blood glucose levels were significantly decreased in resveratrol treatment groups( P〈0. 05),especially in high dose group. The results of HE staining showed that the inner retina was rough and thinning in db/db group,and it was improved significantly in resveratrol groups. Visual electrophysiological test results showed that compared with db/db group,the latency of a wave and b wave was shortened in dark adaptation 3. 0 reaction in resveratrol groups( P〈0. 05). Western blot results showed that the expression of VEGF protein in db/db group was significantly increased compared with control group,but the expression of VEGF was significantly decreased in resveratrol groups.( P〈0. 05) Conclusion Resveratrol can reduce the blood glucose and improve the retinal function in db/db mice,which may be related to the decrease of expression of VEGF protein.
