铁皮石斛(Dendrobium officinale)是中国的珍贵中药材,目前,铁皮石斛野生资源稀缺,组织培养是铁皮石斛快速繁殖的有效途径。近年来,铁皮石斛组织培养上的研究主要有种子无菌萌发,外植体诱导原球茎发生、增殖与分化,在培养基上生根与壮苗等,大量研究表明,以铁皮石斛茎段为外植体进行组培更易获得试管生根苗。原球茎增殖与分化最适基础培养基是MS,NAA浓度为0.5 mg/L时,铁皮石斛生根率为100%,且长势良好,生长健壮。配以一定的添加物能使之更快形成幼苗。同时,幼苗壮苗以MS+IBA 0.5 mg/L+NAA 0.5 mg/L培养基为最佳,茎粗、苗高、叶长,效果最好。但是铁皮石斛组织培养周期长是其快速繁殖中存在的主要问题,对此,建议可以通过遗传变异、单倍体诱导、离体开花授粉等非常规育种方式,快速培育优良品种,或者在进行组织培养研究时,直接以组织培养物替代原药材使用或者生产活性成分等方面进行研究。

At present, the wild Dendrobium officinale resources are scarce.The use of tissue culture is an effective way for rapid propagation of Dendrobium officinde.In recent years,research on the tissue culture of Dendrobium officinale mainly seed germination,protocorm occurrence,proliferation and differentiation of explant in culture medium, rooting and seedling etc. A lot of experimental research showed that, the dendrobium stem segments were used as explants for tissue culture to obtain seedling. Protocorm proliferation and differentiation of the optimum of culture medium was MS,NAA concentration was 0.5 mg/L,Dendrobium officinale rooting rate was 100%,and grown well and robust growth. And with certain additives can make faster formation of seedlings.At the same time,the seedling on MS+IBA 0.5 mg/L+NAA 0.5 mg/L medium was the best,stem diameter,seedling height, leaf length had the best effect. But of Dendrobium officinale tissue culture long period is the rapid propagation in the presence of main problems,therefore,it was suggested by genetic variation,haploid induction ,in vitro flowering pollination breeding way,fast breeding excellent varieties ,or in tissue culture studies,directly in tissue cultures of alternative raw material to use or produce active components in aspects of research.
