抗癌生物活性肽对实验性胃癌PCNA、caspase-8基因表达的影响

目的:探讨抗癌生物活性肽作用于荷胃癌裸鼠后,对其增殖及凋亡相关基因表达变化的影响。方法:将裸鼠接种胃癌MGC-803细胞后使之致瘤。实验随机分为3组,对照组11只,脾肽实验组12只,肝肽实验组11只;分别尾静脉注射生理盐水0.4mL、脾肽0.4mL(0.15mg/mL)、肝肽0.4mL(0.05mg/mL)。连续处理35d,停药4d后处死裸鼠,取肿瘤,以4%多聚甲醛固定,常规石蜡包埋,4μm切片。应用免疫组化SABC法检测PCNA、caspase-8蛋白在实验组与对照组中的表达情况。结果:PCNA蛋白在对照组(NS组)肿瘤中阳性表达较强,在P组、G组裸鼠肿瘤中PCNA蛋白的表达均比对照组的表达弱,且有显著性差异(P〈0.01);caspas-8蛋白在对照组(NS组)肿瘤中阳性表达较弱,P、G组裸鼠肿瘤中caspas-8蛋白的表达均比对照组表达强,且有显著性差异(P〈0.01)。结论:抗癌生物活性肽能降低胃癌移植瘤细胞的增殖活性,并能促进其凋亡。

英文摘要:

Objective:To investigate the expression changes of proliferating and apoptotic related genes of anti-cancer bioactive peptide treated tumor-loaded nude mice.Methods:Gastric cancer cell line MGC-803 was inoculated to nude mice to make them tumor-loaded.Nude mice were divided into 3 groups randomly:11 as control group,12 as spleen peptide group and 11 as liver peptide group.3 groups of nude mice were administered with 0.4mL normal saline,0.4mL spleen peptide(0.15mg/mL)and 0.4mL liver peptide(0.05mg/mL)continuously for 35 days,respectively.Nude mice were executed after the administration finished 4 days later,tumors were taken out and fixed with 4% paraformaldehyde,routine paraffin embedded and sliced into 4μm.Then,PCNA and Caspase-8 expression were detected using immunohistochemistry SABC method between experimental and control groups.Results:The expression of PCNA was relatively stronger in control group than those in experimental groups which showed significant difference(P〈0.01).While the expression of Caspase-8 was relatively weaker in control group than that in experimental groups and which showed significant difference(P〈0.01).Conclusion:Anti-cancer bioactive peptide can inhibit the proliferating activity of transplanted gastric cancer cells and promote cell apoptosis.
