目的 18^F-FDG PET/CT显像评价肝癌H22模型及三萜类中药成分熊果酸(UA)对移植性肝癌(H22)细胞体内生长的抑制作用。方法昆明小鼠20只,雌雄各半,体重(20±2)g,右前腋皮下接种1×107/0.2 ml小鼠肝癌H22细胞,左前腋皮下接种等量生理盐水作为对照。建立实体瘤模型并进行PET/CT检查评价实体瘤模型后,随机分两组,对照组和熊果酸组(UA组),每组10只,UA组每只按4.536 mg/kg给予体积0.2 ml的UA,每天1次,连续10 d;对照组给相同容积的生理盐水。给药结束后再次行18^F-FDG PET/CT显像。显像后处死小鼠,采用HE染色法检测形态学改变。结果 (1)20只小鼠治疗前右前腋皮下肿瘤接种部位放射性摄取/同侧肺(T右1/NT右1)为2.833±0.18,左前腋皮下接种等量生理盐水区放射性摄取/同侧肺(T左1/NT左1)为1.078±0.27,T右1/NT右1与T左1/NT左1有统计学差异(t=15.55,P=0.0000)。(2)对照组与UA组右前腋皮下接种肿瘤部位放射性摄取/同侧肺(T右2/NT右2与T右3/NT右3)分别为2.824±0.224及1.534±0.429,二者有统计学差异(t=11.85,P=0.0001)。左前腋皮下同等体积生理盐水接种放射性摄取/同侧肺(T左2/NT左2与T左3/NT左3)分别为1.078±0.33及1.076±0.34,二者无统计学差异(t=2.121,P=0.078)。结论18^F-FDG PET/CT显像是评价肝癌H22模型及熊果酸抑制肝癌效果的准确方法。
Objective To evaluate model of HepA-ascitic type(H22)cell and the inhibiting effect of ursolic acid(UA)for H22 cell in vivo by 18F-FDG PET/CT imaging.Methods Kunming mice[n=20,10 males,10 females,(20±2)g]were translated by hypodermic(injection)with 1×107/0.2 ml H22 cell to right armpit and translated by hypodermic(injection)with same volume 0.9%NS to left armpit respectively and solid tumor model were established.It divided UA and control group after FDG PET/CT imaging,10 mice were in every group.About 0.2 ml UA(4.536 mg/kg)were pour into stomach of UA group from the first day to the tenth day.Control group were pour same volume 0.9%NS into stomach from the first day to the tenth day.In the eleventh day,the animals performed FDG PET/CT imaging again.Mice were executed after PET/CT imaging and the morphology was observed by HE.Results(1)Radioactivity uptake regions of translated tumor in right armpit vs.same side lung(TR1/NTR1)was 2.833±0.18,Radioactivity uptake regions of translated same volume 0.9%NS in left armpit vs.same side lung(TL1/NTL1)was 1.078±0.27 in 20 mice,TR1/NTR1 vs.TL1/NTL1 was significantly(P〈0.05).(2)Radioactivity uptake regions of translated tumor in right armpit vs.same side lung(TR2/NTR2 vs.TR3/NTR3)by 18F-FDG PET/CT imaging were 2.824±0.224 and 1.534±0.429 in UA and control group respectively,between UA and control group had significant difference(P〈0.05).Radioactivity uptake regions of translated same volume 0.9%NS in left armpit vs.same side lung(TL2/NTL2 vs.TL3/NTL3)were 1.078±0.33 and 1.076±0.34 in UA and control group respectively,between UA and control group had no significant difference(P〈0.05).Conclusions 18F-FDG PET/CT imaging is an accurate and available method to evaluate model of HepA-ascitic type(H22)cell and the inhibiting effect of UA for H22 cell in vivo.
