目的探讨干细胞因子(SCF)对高糖环境下豚鼠膀胱Cajal样间质细胞(ICCs-like)形态及电生理的影响。方法胶原酶消化法原代分离培养豚鼠膀胱ICCs样细胞。实验分组:正常对照组(葡萄糖浓度为5 mmol/L)、高糖组(葡萄糖浓度为15 mmol/L),培养基培养72 h后高糖组分为高糖对照组、SCF 50 ng/mL组、SCF 100 ng/mL组,继续培养24 h及72 h。激光共聚焦显微镜下测量细胞长度并获得每个时间点细胞内钙离子荧光强度值。结果随着培养时间延长,高糖对照组较正常对照组细胞长度明显缩短、钙离子荧光值显著降低(P〈0.01)。SCF 50 ng/mL作用72 h后钙离子荧光值较高糖对照组增高(P〈0.05)。SCF 100 ng/mL作用24 h后较高糖对照组细胞长度增长、钙离子荧光值增高(P〈0.05),而作用72 h后细胞长度明显增长、钙离子荧光值显著增高(P〈0.01)。结论外源性SCF干预后对高糖环境造成的ICCs样细胞形态及电生理功能异常有一定的改善或逆转作用。

英文摘要:

Objective To investigate the effects of stem cell factor(SCF) on the morphology and electrophysiology of interstitial cells of Cajal-like cells(ICCs-like) in the bladder of guinea pigs in high glucose environment.Methods ICCs-like cells were isolated from the bladder of guinea pigs by collagenase digestion,and divided into the normal control group(glucose concentration 5 mmol/L) and high glucose group(glucose concentration 15 mmol/L).After 72 h of culture,the high glucose group was divided into the high glucose control group,SCF 50 ng/mL group and SCF 100 ng/mL group,which were then continued to be cultivated for 24 h and 72 h.The cell length was measured and the fluorescence intensity of calcium was observed by laser scanning confocal microscope.Results With the increase of culture time,the cell length was apparently shorter and the fluorescence intensity of calcium was significantly lower in the high glucose control group compared to that of the normal control group(P0.01).After 72 h of culture,the fluorescence intensity of calcium was higher in the SCF50 ng/mL group than in the high glucose control group(P0.05).The cell length increased and the fluorescence intensity of calcium was higher in the SCF 100 ng/mL group than in the high glucose control group(P0.05) after 24 h of culture.This improvement was more evident after 72 h(P0.01).Conclusions Exogenous SCF may improve the morphology and electrophysiology of ICCs-like cells in the high glucose environment.
