为探讨豚鼠膀胱Cajal样间质细胞(Interstitial cells of Cajal-like,ICCs)的原代培养及鉴定方法。方法:采用胶原酶消化法,得到原代培养的成年豚鼠膀胱ICCs细胞,应用免疫荧光法完成对ICCs的细胞鉴定。采用膜片钳技术,在全细胞记录模式下,记录正常对照组(体外培养72 h,葡萄糖浓度为5 mmol/L;72 h,G 5 mmol/L)、Ih电流鉴定组(体外培养72 h,葡萄糖浓度为5 mmol/L,检测时使用含CsCl 2 mmol/L的细胞外液;72 h,G 5 mmol/L,CsCl2 mmol/L)中ICCs细胞超极化激活时的Ih电流(hyperpolarization activated inward current,Ih)值。结果显示,正常对照组Ih电流数值(505.75±157.66)PA,Ih电流鉴定组(149.89±62.84)PA,正常对照组与高糖Ⅰ组比较P〈0.01。由此可知,用酶消化法可获得豚鼠膀胱Cajal间质细胞,在体外高糖环境培养基中生长良好并能检测出Ih电流。
Objective:To explore the primary cultivation and identification method of the adult guinea pig bladder's ICCs in the high concentrations of glucose environment.Methods:Primarily cultured bladder ICCs of adult guinea pigs were harvested by collagenase digestion.By using immunofluorescence method,cell identification was made.We employed the whole-cell patch clamp technique to record the Ih current values of the ICCs in normal control group and Ih current identification group.Results: The Ih current value was 505.75±157.66 PA in the Nnormal control group,149.89 + 62.84 pA in Ih current identification group(P0.01).Conclusion:The enzymatic digestion is available to cultivate ICCs in guinea pig bladder,the cells grow well in the high concentrations of glucose medium,and Ih current can be detected.
