目的:观察体外高糖环境下Cajal样细胞(interstitial cells of Cajal-like,ICCs)c-kit mRNA表达变化。方法:胶原酶消化法原代分离培养豚鼠膀胱ICCs,加入含葡萄糖浓度分别为5mmol/L、15mmol/L、30mmol/L培养基培养24h、48h、72h后,应用逆转录-聚合酶链式反应(RT-PCR)法检测高糖环境下c-kit基因mRNA表达水平的变化。结果:RT-PCR电泳经分析显示15mmol/L、30mmol/L组ICCs c-kit mRNA表达较5mmol/L组降低(P〈0.01),72h组较24h组、48h组表达降低。结论:高糖环境造成ICCs c-kit基因mRNA表达水平降低,可导致ICCs的SCF/c-kit信号通路异常,ICCs功能及结构障碍,可能是糖尿病膀胱病变的发病机制之一。
Objective: To examine the expressions of interstitial cells of Cajal-like(ICCs) of c-kit mRNA in high glucose in vitro.Methods:The V-type enzyme collagenase was used to separate primary cells from myoblast of the preparation muscle tissue;put the glucose into culture media for cells culture(5mmol/L,15mmol/L,30mmol/L),expression of c-kit mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR) analysis after 24h,48h,72h by cultured.Results:Compared with those of controls,decreased expression of interstitial cells of Cajal-like(ICCs) of c-kit mRNA was demonstrated in the group 15mmol/L、group 30mmol/L(P〈0.01),the change will be more obviously found by degrees of time.Conclusion:Significant down regulation of c-kit gene mRNA in high glucose environment of interstitial cells of Cajal-like,abnormalities expression of c-kit gene at transcription and translation phase may disturb the SCF/c-kit signal pathway to result in obstacle of functions and structure,it may be one of the pathogenesis in diabetes cystopathy.
