以金线莲的茎片、带节茎段和不定芽为外植体,研究不同植物生长激素组合和浓度配比对福建金线莲原球茎诱导、丛生芽诱导及幼苗生根壮苗的影响。结果表明:适合原球茎诱导的培养基为MS+6-BA2.0mg/L+2,4-D0.2mg/L+ZT0.2mg/L+蔗糖30g/L+琼脂7g/L,诱导率达81%;适合丛生芽增殖的培养基为MS+6-BA2.0mg/L+NAA0.5mg/L+TDZ0.2mg/L+蔗糖30g/L+琼脂7g/L.丛生芽诱导增殖效果最佳,90d时增殖倍数达23倍;适合不定芽生根的培养基为1/2MS+NAA0.5mg/L+IBA4.0111g,L+6-BA0.1mg/L+蔗糖30∥L+琼脂7g/L+AC3g/L+香焦汁100g/L。
Effects of different hormone on tissue culture of Anoectochilus roburghii were studied using stem tablets, stem segments and young shoots as explants respectively. The results indicated that the optimized medium of protocorm-like bodies (PLB) induction was MS+6-BA 2.0 mg/L+2,4-D 0.2 mg/L+ZT 0.2 mg/L+sucrose 30 g/L+agar 7 g/L; the appropriate medium for induction and proliferation of adventitious bud was MS +6-BA 2.0 mg/L+NAA 0.5 mg/L+TDZ 0.2 mg/L +sucrose 30 g/L +agar 7 g/L and the proliferation rate reached 23-fold after 90 days. The best rooting medium was 1/2MS+NAA 0.5 mg/L+IBA 4.0 mg/L+6-BA 0.1 mg/L+ sucrose 30 g/L+sugar 7 g/L+AC 3 g/L+banana juice 100 g/L.
