目的:建立测定壳聚糖微球中葛根素载药率的高效液相色谱测定法。方法:微球经盐酸回流降解并用蒸馏水定容后进行测定。葛根素的定量分析采用Eclipse XDB-C18色谱柱(4.6 mm×150 mm,5μm),流动相为甲醇-水(20∶80),流速1 mL.min^-1,检测波长250 nm,柱温30℃,进样量为20μL。结果:在本文确定的色谱条件下,葛根素浓度在0.2~20.0μg.mL^-1范围内线性关系良好(r=0.9998,n=6);平均回收率(n=9)为100.3%。结论:本方法可用于壳聚糖微球中葛根素的载药率测定。
Objective:To establish a method for determination of the loading efficiency of puerarin in chitosan microspheres.Methods:The microspheres were degraded by hydrochloric acid solution,and the drug content could be measured after being diluted with distilled water.The quantitative analysis of puerarin was carried out by HPLC with Eclipse XDB-C18 column(4.6 mm×150 mm,5 μm),the mobile phase was methanol-water(20∶ 80) at a flow rate of 1.0 mL·min^-1,the detection wavelength was 250 nm,the column temperature was 30 ℃ and the injection volume was 20 μL.Results:Using the chromatographic condition in this paper,puerarin had a good linear relationship in the range of 0.2-20.0 μg·mL^-1(r=0.9998,n=6),the average recovery(n=9) was 100.3%.Conclusion:The method can be used for quality control of puerarin in chitosan microspheres.
