大孔吸附树脂分离纯化补阳还五汤中总黄酮

目的:探究大孔吸附树脂法分离纯化补阳还五汤中总黄酮的最佳工艺条件和参数。方法:通过静态吸附及解吸试验、动态吸附及洗脱试验,从D101,DA201,AB-8和聚酰胺树脂中筛选出适合补阳还五汤总黄酮分离的理想树脂。并通过单因素分析考察该树脂分离、纯化补阳还五汤总黄酮的最佳工艺条件。结果:经D101大孔吸附树脂纯化后的补阳还五汤总黄酮纯度最高,达到63.1%,转移率为80.2%,静态吸附容量为66.28 mg·g-1(干树脂),最佳动态上样量为20 mL·g-1(总黄酮质量浓度:3.033 g·L-1),上样流速为1 BV·h-1,洗脱乙醇体积分数为60%,洗脱速度为1.5 BV·h-1。结论:D101大孔吸附树脂可较好地用于补阳还五汤总黄酮的分离纯化。

英文摘要:

Objective:To investigate the optimum conditions and parameters of macro reticular resin in purification of flavonoids in BYHWD.Method:Through the static adsorption and desorption experiments and the dynamic adsorption and desorption experiments,the ideal resins for BYHWD flavonoids from the D101,DA201,AB-8 and polyamide were screened.Through single-factor analysis to study the optimum conditions of resin in separating and purifying BWD flavonoids.Result:After purification by D101,BYHWD the purity of flavonoids could reach 63.1%,transfer rate was 80.2%.The static adsorption capacity of D101 macroporous resin for BYHWD flavonoids was 66.28 mg.g-1(dry resin),the best dynamics on the sample size was 20 mL.g-1(total flavonoids concentration 3.033 mg.mL-1),the sample flow rate was 1 BV.h-1,elution ethanol concentration was 60%,elution speed was 1.5 BV.h-1.Conclusion: D101 macroreticular resin could be better used for the separation and purification of BWD flavonoids.
