目的:探讨荧光分光光度法测定当归中阿魏酸含量的方法。方法:以2%碳酸钠为溶剂,在λex为343nm、λem为463nm处测定当归的荧光强度。结果:阿魏酸含量在0.3-3μg/mL范围内呈良好的线性关系,线性回归方程为y=19.469x+4.8058,r=0.997,阿魏酸对照品在3h内稳定,表明该法灵敏度高、重现性较好,且操作简便。结论:荧光分光光度法可以用于当归药材中阿魏酸含量的测定,且方法简单准确、灵敏度高。
Objective:To establish a new method for the contents determination of ferulic acid in Radix angelicae sinensis based on fluorescence spectrometry.Methods:The fluorescence intensity of Radix angelicae sinensis dissolved in 2% sodium carbonate was determined at the excitation wavelength of 343 nm and the emission wavelength of 463 nm.Results:The linear range of ferulic acid was 0.3-2.2 μg/mL,the linear regression equation was y = 19.469x +4.805 8 with relationship coefficient of 0.997.Ferulic acid reference substance was stable within 3 hours.The results showed that the method was rapid,sensitive and repeatable.Conclusions:Fluorescence spectrometric method can be used to determine ferulic acid contents in Radix angelicae sinensis for its simpleness,precise and high sensitivity.
