目的:建立土炒当归饮片质量控制方法,为制定土炒当归饮片质量标准提供参考。方法:按照2005年版《中国药典》规定,对土炒当归饮片的性状、显微鉴别、水分、灰分、酸不溶性灰分、浸出物和薄层色谱进行分析,并采用高效液相指纹图谱法对其化学成分及相对含量进行研究。结果:土炒当归表面微挂土色,符合当归饮片土炒的传统炮制标准;其水分、总灰分、酸不溶性灰分和浸出物的平均含量分别为5.8%、14.6%、6.9%和49.7%,但不同产地当归有明显差异,土炒后灰分和酸不溶性灰分较未炮制前有所增加;建立了不同产地土炒饮片的化学指纹图谱,确定了11个共有峰,其相对含量因产地不同虽略有差异,但色谱图概貌一致,相似度均大于90%。结论:根据《中国药典》规定,并结合高效液相指纹图谱法建立了土炒当归质量评价标准,为土炒当归饮片的生产、经营及鉴定提供参考标准。
Objictive:To establish the quality standard of soil-processed decoction pieces of angelica.Methods:Characters,microstructure,water contents,general ash,acid insoluble ash content,extract contents and thin layer chromatography of soil-processed angelica were detected referring to China pharmacopoeia (2005).On this basis,chemical compositions and their relative contents were analyzed using high performance liquid chromatography (HPLC) fingerprint.Results:The surface of soil-processed decoction pieces present soil color,which accord with traditional processed standard;the average contents of water,general ash,acid insoluble ash and extract was 5.8%,14.9%,6.9% and 49.7% respectively,which was significantly different between different producing areas;contents of general ash and acid insoluble ash in soil-processed decoction pieces were higher than that in non-processed ones.A chemical fingerprint method was developed and 11 common peaks were found in all soil-processed decoction pieces of angelica.The content or relative content of 11 common peaks was slightly different between different producing areas.However,chromatographic patterns of all samples were generally consistent and similarities of all samples were higher than 90%.Conclusion:The quality standards of soil-processed angelica was established using the method of China pharmacopoeia (2005) and HPLC fingerprint,as a reference for identification,management and production of soil-processed decoction pieces of angelica.
