本文报道了酸性柴胡多糖(WBP011)在各种环境中的聚集态行为。利用热分析系统改变酸性柴胡多糖(WBP011)的环境温度,跟踪检测到该多糖的聚集态特性的变化,表明在不同温度环境下,该多糖有多种聚集形态。利用原子力显微镜观察云母片表面WBP011分子的形貌,研究不同溶剂对该多糖分子及聚集体行为的影响。通过观察发现,水溶液中WBP011分子呈旋绕态的柔性链;盐离子的浓度在一定程度上控制着多糖分子构象的变化;碱溶液中由于分子间负电荷数目的增加强化了静电排斥作用,使WBP011分子呈刚性排列,与在水溶液中的排列有明显不同。以上结果表明WBP011分子构象的变化依赖于WBP011/云母片界面及WBP011分子/WBP011分子间的相互作用。
Aggregation behavior of acidic Bulpeurum Polysaccharide (WBP011) in a variety of environments was investigated. Using thermal analysis to trace the behavior of aggregation of polysaccharide with temperature rising, it was discovered that the change of the configuration of acidic Bulpeurum Polysaccharide (WBP011) in different temperature. The effect of different solvents on aggregation behavior of acidic Bulpeurum Polysaccharides was investigated using atomic force microscope observing WBP011 molecular morphology on the surface of mica in Atmospheric environment. The concentration of ions controlled conformation changes of acidic Bulpeurum Polysaccharide (WBP011) in a certain extent. In alkali solution, the increasing of negative charge strengthened the role of the electrostatic repulsion, consequently WBP011 molecules exhibited rigid structures. The above results showed that conformation of WBP011 molecular changed dependently on the interfacial change between WBP011 and mica, as well as interaction among WBP011 moleeulars.
