目的 探索依达拉奉对脑缺血再灌注大鼠脑红蛋白(Ngb)和14-3-3γ蛋白的影响.方法 采用大脑中动脉闭塞法建立大鼠脑缺血再灌注模型,将48只雄性SD大鼠随机分为假手术组、模型组和依达拉奉预处理组;采用酶联免疫吸附法检测血清Ngb和14-3-3γ水平,采用蛋白免疫印迹、免疫组化检测脑组织中Ngb和14-3-3γ表达.结果 模型组大脑和血清的Ngb和14-3-3γ水平均高于假手术组(P〈0.05),依达拉奉预处理组大脑和血清的Ngb和14-3-3γ水平均高于模型组(P〈0.05).结论 依达拉奉能通过上调脑缺血再灌注大鼠血清和脑组织中Ngb和14-3-3γ水平发挥脑保护作用.
Objective To explore the effect of edaravone on neuroglobin(Ngb) and 14-3-3γ protein in rats with middle cerebral artery ischemia-reperfusion(I/R).Methods Rat model of I/R and reperfusion were established by the cerebral artery occlusion method.48 male SD rats were randomly divided into sham operation,model and edaravone-pretreated groups.The expressions of Ngb and 14-3-3γ were detected by enzyme-linked immunosorbent assay (ELISA),immunohistochemistry and Western blot.Results The Ngb and 14-3-3γ levels of brain and serum in model group were higher than those of sham operation group (P〈0.05),while Ngb and 14-3-3γ expressions were higher in edaravone-pretreated group compared to those of model group (P〈0.05).Conclusions Edaravone has a protective effect against middle cerebral artery I/R by enhancing Ngb and 14-3-3γ.
