目的探讨依达拉奉对脑缺血再灌注大鼠脑组织和血清血管细胞黏附分子1(VCAM-1)和细胞间黏附分子1(ICAM-1)水平的影响。方法将30只雄性SD小鼠随机分为假手术组、缺血再灌注组和依达拉奉组,每组10只。建立大鼠大脑中动脉缺血再灌注模型,24h后采集脑组织和血清,分别采用免疫组织化学、ELISA法检测VCAM-1和ICAM-1水平。结果与假手术组比较,缺血再灌注组大鼠脑组织和血清ICAM-1、VCAM-1明显升高(P〈0.05);与缺血再灌注组比较,依达拉奉组脑组织ICAM-1、VCAM-1[(0.14±0.02)μg/L vs(0.19±0.04)μg/L、(0.12±0.02)μg/L vs(0.17±0.03)μg/L,P〈0.05]和血清ICAM-1、VCAM-1[(1.03±0.29)μg/L vs(1.29±0.44)μg/L、(170.79±43.42)μg/L vs(261.85±73.05)μg/L,P〈0.05]明显降低。结论依达拉奉能改善脑缺血再灌注大鼠的症状,其作用机制之一是抑制ICAM-1和VCAM-1的生成。
Objective To study the effect of edaravone on serum VCAM-1and ICAM-1levels in rats following I/R injury.Methods Thirty male SD rats were randomly divided into sham operation group,I/R injury group and edaravone treatment group(10in each group).Twenty-four hours after a rat cerebral middle artery I/R injury model was established,brain tissue and serum samples were taken.The serum VCAM-1and ICAM-1levels were measured by enzyme-link immunosorbent assay with immunohistochemical staining.Results The serum ICAM-1and VCAM-1levels were significantly higher in I/R injury group than in sham operation group(P〈0.05)and significantly lower in edaravone treatment group than in I/R injury group(1.03±0.29μg/L vs1.29±0.44μg/L,170.29±43.42μg/L vs 261.85±73.05μg/L,P〈0.05).Conclusion Edaravone improves the symptoms of cerebral I/R injury in rats by inhibiting the formation of VCAM-1and ICAM-1.
