采用基因克隆方法首次扩增出黑线仓鼠催乳素受体(Prolactin Receptor,PRLR)基因5’端部分序列(GenBank登录号为EU826030),测序结果表明该片段的有效长度为379bp,包含部分5’调控区、exonl及部分intron1.该序列与其它物种相应区域的同源性比较结果:黑线仓鼠与人、大鼠、小鼠、狐猿、黑猩猩、类人猿PRLR基因核苷酸序列的同源性为82.1%-93.4%;exon1序列的同源性为79.8%-91.4%.系统进化分析与物种亲缘关系的远近一致,同时证明PRLR基因exon1有较大的可变性,因此认为成功克隆了黑线仓鼠的PRLR基因的部分序列.该研究所得到的PRLR基因序列可用作研究物种亲缘关系或遗传距离的理想标记.
The partial sequence of 5'end of prolactin receptor(PRLR) gene in Cricetulus barabensis was first cloned in this experiment, with Genbank submittion number EU826030. The 379 bp virtual fragment which contained partial sequence of 5'end, the full length of exon 1 and partial intronlwas amplified successfully by PCR. The nucleotide homology of PRLR gene among Cricetulus barabensis ,Rattus norvegicus, Mus musculus, Homo sapiens, Pongo abelii, Macaca mulatta and Pan troglodytes was from 82.1% to 93.4%. The homology of exonl was from 79.8% to 91.4%. This result was similar to former reports, what's more, it could prove that the exonl was variable. So the partial fragment of Cricetulus barabensis was cloned successfully. To this content, it could be considered as an ideal marker for studying relationships among animals and genetic distance.
