目的:研究转染重组Elafin对炎症状态下巨噬细胞表面受体CD14的保护作用。方法:重组质粒pEG-FP-C1-Elafin转染入离体培养巨噬细胞株,与多形核中性粒细胞(polymorphonuclear granulocyte,PMN)、脂多糖(li-popolysaccharide,LPS)共作用4 h后,倒置荧光显微镜观察重组Elafin胞内表达,流式细胞仪检测巨噬细胞表面受体CD14表达情况。同时,转染重组质粒的巨噬细胞与凋亡PMN共孵育1 h后,观测结合凋亡PMN的巨噬细胞百分比。结果:pEGFP-C1-Elafin转染入巨噬细胞后被成功表达,其表面受体CD14数量明显升高,结合凋亡PMN的巨噬细胞的数量的增加。结论:通过转染重组人Elafin,可保护炎症状态下巨噬细胞表面凋亡相关受体CD14,促进PMN凋亡。
Aim:To investigate protective effect of recombinant Elafin on CD14 receptor of macrophage challenged by polymorphonuclear granulocyte(PMN) and lipopolysaccharide(LPS).Methods: Plasmid pEGFP-C1-Elafin was transfected into macrophage,then challenged for 4h by PMN and LPS.Expressed Elafin was detected by inverted fluorescence microscope and CD14 was determined by flow cytometry.Transfected macrophage cocultured for 1h with apoptotic PMN to observe the number of macrophage interacted with apoptotic PMN.Results: Transfected pEGFP-C1-Elafin was expressed successfully in macrophage,of which the CD14 quantity increased in evidence.And the amount of macrophage combined with apoptic PMN was also elevated.Conclusion:The approach of genetic modification of macrophage with Elafin can be used to restore apoptotic related receptor CD14 in inflammatory condition.
