目的:研究白细胞介素-6(IL-6)和白细胞介素-18(IL-18)在复发性流产(RSA)患者绒毛组织中的表达,探讨RSA的发病机制。方法采用实时荧光定量逆转录聚合酶链反应技术,检测63例RSA妇女(RSA组)及50例正常早孕妇女(正常组)绒毛组织中IL-6mRNA和IL-18mRNA的表达水平,并将两者进行相关性分析。结果RSA组绒毛组织中IL-6mRNA的表达水平显著低于正常组,IL-18mRNA水平显著高于正常组,相关分析显示,RSA组IL-6与IL-18mRNA两者之间表达水平呈负相关(r=-0.49,P〈0.01)。IL-6mR—NA的表达水平与RSA患者流产次数无关,而IL-18mRNA表达水平随着流产次数增多而增加,差异有统计学意义(P〈0.05)。结论正常妊娠较RSA患者绒毛组织IL-6水平高,当IL-6表达降低或IL-18升高时,可能导致RSA,其中IL-18水平与流产次数呈正相关。
Objective To investigate the expression of interleukin -6 (IL -6) and interleukin -18 (IL - 18) mRNA in villi of patients with recurrent spontaneous abortion (RSA) and to investigate the pathogenesis of RSA. Methods Real - time quantitative reverse transcription - polymerase chain reaction was used to quantify the expression of IL - 6 mRNA and IL -18 mRNA in villi of 63 patients with RSA (RSA group) and 50 normal early pregnant women( normal group). The correlation between IL- 6 and IL- 18 was analyzed. Results IL- 6 mRNA was significantly lower, while IL - 18 mRNA was significantly higher, in RSA group than that in normal group. Meanwhile, there was significantly nega- tive correlation between the IL- 6 and IL- 18 in RSA group (r = -0. 49, P 〈0. 01 ). Although there was no significant correlation between the number of abortion and IL- 6 mRNA, IL- 18 mRNA was significantly elevated with the increase of number of abortion ( P 〈 0. 05 ). Conclusion The down - regulated IL - 6 or up - regulated IL - 18 can lead to RSA, as IL - 18 is furthermore positively correlated with the number of abortion.
