目的:探讨湖南地区人群早发冠心病(CHD)血瘀证与血管紧张素原(angiotensinogen,AGT)基因Rs4762和Rs699多态性的关系。方法:通过基于飞行时间质谱分析技术(TOF-MS)和等位基因特异性PCR技术方法,从湖南地区人群中选取早发冠心病血瘀证患者50例,早发冠心病非血瘀证患者26例,非早发冠心病患者36例和健康对照组19例,对AGT基因Rs4762(T174M/C3889T)和Rs699(M235T/T4072C)多态性位点进行检测,分析四组等位基因频率以及基因型频率的差异性。结果:Rs4762、Rs699多态位点的等位基因在湖南地区人群中早发冠心病血瘀证组与健康对照组之间存在显著差异性(χ^2=14.872,P〈0.01;χ^2=9.512,P〈0.01),Rs4762、Rs699多态位点的基因型在湖南地区人群中早发冠心病血瘀证组与健康对照组之间也存在差异性(χ^2=13.371,P〈0.01;χ^2=7.889,P〈0.05)。结论:AGT基因Rs4762、Rs699多态性与湖南地区人群早发冠心病血瘀证均有相关性,Rs4762多态位点和Rs699多态位点CC型基因是湖南地区人群早发冠心病血瘀证的易感基因。
Objective:Discussion population with premature coronary heart disease in Hunan(CHD)blood stasis and AGT gene polymorphisms Rs699 Rs4762 and relationships. Methods:The specific PCR technology and technology-based flight mass spectrometry(TOF-MS)method to select patients with premature coronary heart disease and blood stasis syndrome 50 cases from the crowd in Hunan Province,early-onset non-BSS 26 cases of patients with coronary heart disease,early-onset coronary heart disease and 36 cases of non-healthy control group of 19 patients,for AGT gene Rs4762(T174M/C3889T)and Rs699(M235T/T4072C)polymorphisms were detected,analyzed four groups of allele frequencies and genotype frequency differences. Results:Rs4762,Rs699 alleles of polymorphic loci in Hunan Province early onset crowd there are significant differences between the CHD group and the healthy control group(χ^2= 14.872,P=0.001;χ^2=9.512,P=0.002),Rs4762,there are also differences(χ^2=13.371,P=0.001;χ^2=7.889,P=0.019)between the genotypes of polymorphic loci Rs699 premature coronary heart disease group and the healthy control group in Hunan province crowd. Conclusion:AGT gene Rs4762,Rs699 Hunan region polymorphism and premature coronary heart disease and blood stasis syndrome populations were correlated,Rs4762 and Rs699 polymorphic loci polymorphic loci CC genotype Hunan population with premature coronary heart disease and blood stasis permit susceptibility genes.
