采用两种不同的介质组合对三色革裥菌(Lenzites tricolor)胞外漆酶进行分离纯化,第一种组合方式为:(NH4)2S04盐析,透析,聚乙二醇(PEG20000)浓缩,SephadexG-150柱层析(1.6cm×60cm),DEAE-Sepha-dexA-25离子交换层析(2.6cm×40cm);第二种组合方式为:(NH4)2S04盐析、透析、超滤浓缩、DEAE-Sepha-rose FF离子交换层析(1.6cm×60cm)、SephadexG-75凝胶层析(1.6cm×60cm)。测定结果表明,第二种组合方式较高效、合理。该漆酶反应活力最高时温度为60℃、最适pH值为4.6。所测试浓度下,Fe^2+、Fe^3+、硫脲、叠氮化钠和二硫苏糖醇对漆酶活性有一定抑制作用;Ca^2+、乙二氨四乙酸及其二钠盐对漆酶活性具明显的促进作用。氧化丁香醛连氮的米氏常数Km值为43.4μmol/L。
An experiment was conducted to purify the extracellular laecase from Lenzites tricolor by two sets ot methods. The difference of the two methods reflects in the assembling of different chromatography media. The first set method is ( NH4 ) 2 SO4 salting-out, dialyzing, polyethylene glycol ( PEG 20000) concentration, SephadexG-150 column chromatogra- phy ( 1.6 cm×60.0 cm), DEAE-SephadexA-25 ion exchange chromatography (2.6 cm×40.0 cm), and the second set method is (NH4 )2 SO4 salting-out, dialyzing, ultrafiltration concentrating, DEAE-Sepharose FF ion exchange chromatography ( 1.6 cm×60.0 cm) and SephadexG-75 gel filtration ( 1.6 cm ×60.0 cm). Result shows that the second method is more efficient and scientific. The optimum temperature and pH value for this lacease are 60 degrees C and 4.6 respectively in catalytic reaction of oxidizing syringaldazine. Various factors show different effects on the laccase activity. The activity is restrained partly by Fe^2+, Fe^3+, thiourea, sodium azide and dithiothreitol (DTT), while is promoted evidently by Ca^2+ , ethylene diamine triacetic acid (EDTA) and disodium EDTA. The laccase catalyzes the oxidation of syringaldazine with a Michaelis constant of 43.4 μmol/L.
