目的检测热激对Jurkat细胞中主要组织相容性复合物Ⅱ类转录激活因子(CⅡTA)的表达,及其对下游人类白细胞抗原(HLA-DR)表达的影响。方法采用实时RT-PCR检测42℃热激和干扰素-γ(IFN-γ)作用前后Jurkat细胞中CⅡTA mRNA的变化。采用Western blot检测热激和IFN-γ作用前后Jurkat细胞中CⅡTA蛋白的表达变化。采用流式细胞仪荧光分析热激前后Jurkat细胞表面HLA-DR的表达。结果在Jurkat细胞中,热激可诱导CⅡTAmRNA和蛋白的表达,而IFN-γ处理后无明显变化。与正常Jurkat细胞相比,热激后HLA-DR在Jurkat细胞表面的抗原浓度明显增加(P<0.01)。结论热诱导Jurkat细胞中CⅡTA和HLA-DR先后表达增高。提示热激可能在免疫基因表达缺陷的细胞中,重建相关基因的功能,为肿瘤热疗提供新的依据。
Objective To explore the effect of a non-lethal heat shock,in comparison with the treatment of interferon-gamma(IFNγ),on the expression of major histocompatibility complex Ⅱ transactivator(CⅡTA) and its downstream target gene of the human leukocyte antigens(HLA)-DR in Jurkat cells.Methods The changes of CⅡTA mRNA in Jurkat cells before and after the treatment of heat shock or IFNγ were detected using real time RT-PCR.The changes of CⅡTA protein were detected with Western blot.The expression of HLA-DR was de...
