青藤碱给药后家兔血液及关节液中药物浓度变化的关系

目的利用微透析技术考察青藤碱给药后家兔血液及关节液中药物浓度变化的关系。方法将微透析探针植入家兔血液及膝关节液中,家兔灌胃给药或膝关节处经皮给药或腹部经皮给药50 mg/kg后,收集家兔血液及关节液中的微透析样品,采用HPLC-MS法测定微透析样品中青藤碱的浓度,观察家兔血液及关节液中青藤碱浓度变化的规律。结果不同的给药途径,不同的给药部位,血液和关节液中的青藤碱浓度变化的趋势是相似的,但灌胃给药和腹部经皮给药,血液和关节液中的青藤碱浓度差异有统计学意义(P〈0.05),而膝关节处经皮给药后血液和关节液中的青藤碱浓度差异无统计学意义(P〉0.05)。结论青藤碱给药后血液和关节液中的药物浓度变化趋势是相似的,但浓度高低与给药途径及给药部位有关。

英文摘要:

Objective To study the relationship of sinomenine concentration between the blood and the knee joint flu id in rabbit by microdialysis.Methods The microdialysis probes were implanted in rabbits' vein and knee joint cap sule to collect samples,the sinomenine were given by intragastric administration,transdermal administration on ab domen and transdermal administration on knee joint,the dosages were 50 mg/kg.The concentration of sinomenine in dialysate samples were detected by HPLC-MS,then the change rule of sinomenine concentration in the blood and the knee joint fluid in rabbit were observed.Results The change trend of the sinomenine concentration in blood and knee joint fluid was accordant,no matter what kinds of way to give drug.The concentration was different when the drug was given by intragastric administration and transdermal administration on abdomen(P〈0.05),and there was no difference when the drug was given by transdermal administration on knee joint(P〈0.05).Conclusion The chang trends of the sinomenine concentration in blood and knee joint fluid are similar,the drug concentration has related with the way of delivery and administration place.
