目的建立益髓生血颗粒(山茱萸,何首乌,阿胶等)UPLC色谱指纹图谱,为其质量控制提供可参考依据。方法采用UPLC法,AcquityUPLCBEHC18色谱柱(50mm×2.1mm,1.7μm),柱温30℃,体积流量0.25mL/min,进样量3止,检测波长254nm,乙睛旬.1%磷酸水溶液梯度洗脱。结果用中药色谱指纹图谱相似度评价系统对10批样品的指纹图谱进行峰匹配,标定了16个共有峰,选取的共有峰在单味药材中均找到归属,并确定了其中9个共有峰的化学结构。10批益髓生血颗粒中共有峰的相对保留时间RSD小于1%,指纹图谱的相似度在0.80以上。结论该法建立的指纹图谱灵敏度高,稳定性好,可以作为益髓生血颗粒的质量控制手段并为其有效成分的研究提供依据。
AIM To establish chromatographic fingerprint of Yisui Shengxue Granules ( Corni Fructus, Polygoni multifiori Radix, Asini colla Corii, etc. ) by UPLC method for evaluating the quality of the preparation. METHODS The optimal UPLC conditions for Yisui Shengxue Granules were adopted with Acquity UPLC BEH C18 column (50 mm ×2. 1 mm, 1.7 μm), acetonitrile-0. 1% phosphoric acid as a mobile phase, the flow rate was 0.25 mL/min with DAD detected at 254 nm, the column temperature was 30 ℃. RESULTS Most peaks could be separated preferably, sixteen common peaks were obtained in the fingerprint of ten batches of Yisui Shengxue Granules with the help of peaks match with Similarity Evaluation System for Chromatographic Fingerprint of TCM, and the RSD of the relative relation time of 10 batches of Yisui Shengxue Granules was lower than 1% , the similarity among fingerprints was calculated more than 0. 80. Total of common peaks selected were found belonging to their single medicinal material, and nine compounds were identified among them. CONCLUSION The methodological studies indicate that this UPLC fingerprint is of high precision, strong reproducibility and good stability. The method can be used to evaluate the quality and to provide evidence for the research of effective com- ponents of Yisui Shengxue Granules.
