目的:建立益髓生血颗粒的血清特征图谱,并分析鉴定其体内吸收入血化学成分。方法:取SD大鼠,灌胃给予不同浓度的益髓生血颗粒,分离含药血清,UPLC法测定,建立益髓生血颗粒含药血清的UPLC图谱分析方法;分析益髓生血颗粒提取物及其含药血清特征图谱;比对鉴定血中成分及其代谢产物。结果:随着药物浓度的提高,血中可检出的移行成分的种类增加,含量提高。高浓度益髓生血颗粒灌胃给药后与空白血清相比,多了13个移形成分,其中4个为原型成分,9个为代谢产物,原型成分有来源于补骨脂的补骨脂素、异补骨脂素。结论:血中成分及代谢产物可能为益髓生血颗粒体内直接作用的类药成分,其分别来源于益髓生血颗粒的组成药味。
Objective :To establish the characteristic chromatograms of serum with Yisui Shengxue Granule, and analyze and i- dentify its chemical compositions absorbed into blood. Methods : SD rats were orally given Yisui Shengxue Granule of various con- centrations. The medicinal serum samples were separated and detected with UPLC. The analysis methods of UPLC characteristic chromatograms of Yisui Shengxue Granule and medicinal serum were established. The characteristic chromatograms of the whole formula of Yisui Shengxue Granule and medicinal serum were analyzed, and the chemical compositions, original crude medicinal and metabolites in blood were compared and identified. Results:It showed the drug concentration was increased;the transitionaltype of component that could be detected in blood was increased, increasing the content of component. The types of chemical com- positions in the blood of rat fed high concentration' s Yisui Shengxue granule were more 13 than the types of chemical composi- tions of the control group, among them 4 were original compositions and 9 were metabolites. The results of UPLC showed that the original compositions were kaempferol and isopsoralen, which came from Radix Astragali. Conclusion:The chemical compositions and metabolites in blood may be the drug- like compositions with the directactions in vivo, which come from the ingredients of Yisui Shengxue Granule.
