目的观察颅咽管瘤组织中IFN-α及其受体IFN-αR和骨桥蛋白(OPN)的表达变化,并探讨其意义。方法采用免疫组化法检测58例颅咽管瘤组织(颅咽管瘤组)及12例同期脑肿瘤造瘘切除的正常脑组织标本(对照组)中的IFN-α、IFN-αR、OPN。所有标本进行HE染色以观察其显微结构。结果颅咽管瘤组织可见炎性细胞浸润。颅咽管瘤组IFN-α、IFN-αR、OPN表达均高于对照组(P均〈0.05),且造釉细胞型颅咽管瘤高于鳞状乳头型者;复发颅咽管瘤高于初发者(P均〈0.05)。颅咽管瘤组织中IFN-αd与OPN的表达呈正相关(rs=0.814,P〈0.01)。结论IFN-α、IFN-αR及OPN在颅咽管瘤组织中高表达,三者共同参与了颅咽管瘤的发病,与肿瘤炎症反应程度有关。
Objective To observe expression of IFN-α, IFN-αR, OPN in craniopharyngima and investigate its signifi- cance. Methods Surgical specimens from 58 patients with craniopharyngioma( craniopharyngioma group) and 12 patients underwent cerebral cortex colostomy( control group) were retrieved. After surgical resection, specimens were fixed in 10% neutral buffered formalin and embedded in paraffin blocks. Sections (4-μm thick) were stained with haematoxylin-eosin for histological examination. IFN-α, IFN-αR, OPN in surgical specimens were detected by immunohistochemical staining. Resuits In the craniopharyngioma tissues, inflammatory cell infiltration were evident, but not in the normal brain tissues. It was found that the expression level of IFN-α, IFN-αR, OPN of craniopharyngioma tissues were significantly higher than that in normal brain tissues( all P 〈0.05 ). Expression of IFN--α, IFN-αR, OPN in craniopharyngioma group were higher than control group, which in ameloblastic cell type were higher than the squamous papillary type, and recurrence craniopharyngi- oma tissue were higher than onset craniopharyngioma ( all P 〈 0.05 ). Expression of IFN--α and OPN were positively correlated in craniopharyngioma organization( rs = 0.814, P 〈 0. Ol ). Conclusions IFN-α, IFN-αR, OPN are over-expressed in the craniopharyngiomas. They are well-established risk factors of craniopharyngioma and play an important role in the craniopharyngioma inflammation.
