目的:在藏酋猴中,寻找和克隆与人clock基因类似的基因。方法:提取藏酋猴外周血白细胞RNA,逆转录为全基因组eDNA,利用人clock基因保守序列设计相关引物,克隆,测序,进行生物信息学分析。结果:扩增得到1994bp的核酸序列。该序列与人,苏门答腊猩猩,绵羊,小鼠,褐家鼠,原鸡和蟾蜍clock基因核酸序列同源性分别为99%,98%,94%,90%,90%,82%和75%。结论:藏酋猴基因组中存在与人clock基因相似的基因,该基因可能为藏酋猴clock基因。
Objective: To clone the gene similar to human gene clock from Macaca speciosa thibetanas. Methods:The gene was am- plified by RT-PCR from the peripheral blood RNA of Macaca speciosa thibetanas with primers designed with human gene clock. Resuits:There was a 1994bp nucleotide sequence was amplified from the peripheral blood RNA of the Macaca speciosa thibetanas, which was 99%, 98%, 94%, 90%, 90%, 82% and 75% homology with Homo sapiens, Pongo abelii, Mus musculus, Rattus norvegicus, Gallus gallus and Xenopus, respectively. Conclusion:There is a gene similar to human gene clock, which may be clock gene clock in Macaca speciosa thibetanas.
