以红鳍东方鲀鱼为原料,从其皮中提取酸溶性胶原蛋白(ASC)和酶溶性胶原蛋白(PSC)。在此基础上,分析了红鳍东方鲀胶原蛋白的结构组成和纯度、热变性、氨基酸组成等基本理化性质。结果表明:红鳍东方鲀鱼皮中酸溶性胶原蛋白和酶溶性胶原蛋白纯度分别高达83.19%和85.00%;将所得胶原蛋白分别经紫外光谱分析、傅里叶红外光谱分析和SDS-PAGE电泳测定,确定酸溶性和酶溶性胶原蛋白符合Ⅰ型胶原蛋白的特征,三螺旋结构完整;经测定,ASC和PSC热变性温度分别为29.85和30.10℃。
The acid-solubilized collagen(ASC)and pepsin-solubilized collagen (PSC)extracted from skin of Takifugurubripes were studied on its physicochemical properties, including the structure, purity, thermal denaturation temperature and the amino acid composition, etc. The results show that the ASC and PSC had high purity of 83.19 % and 85.00 %, respectively. The analysis of UV spectra, FTIR spectra and SDS-PAGE of ASC and PSC show that the two kinds of collagen were proved to be proteinⅠwith integrate triple-helix structure. And the thermal denaturation temperatures of ASC and PSC were 29.85 and 30.10 ℃, respectively.
