目的检测银屑病中抑癌基因TIG1的表达变化以揭示其在寻常型银屑病发病机制中的作用。方法用原位杂交的方法检测正常组皮肤组织、银屑病组未受累皮肤组织和皮损中TIG1m RNA的表达。结果在正常组皮肤组织中,TIG1表达于表皮全层;在银屑病组未受累皮肤组织和银屑病边缘皮损中,可见TIG1表达于基底上层,在银屑病中间皮损中无表达。TIG1在银屑病边缘皮损和未受累组织基底层中的表达低于其在正常组皮肤基底层中的表达(P〈0.01);TIG1在银屑病中间皮损基底上层中的表达低于其在未受累皮肤和正常组皮肤的表达(P〈0.01)。结论TIG1可保持表皮正常分化功能,TIG1的降低可能与银屑病角质形成细胞的异常分化相关。
Objective To detect alteration of anti-oncogene TIG1 mRNA in order to reveal the mechanism of TIG1 in psoriasis vulgaris. Methods Expression of TIG1 mRNA in normal tissues, uninvolved skin tissues and psoriatic lesions was detected by in situ hybridization. Results TIG1 mRNA was expressed in all layers of normal epidermis. In uninvolved tissues and the marginal part of psoriatic lesions, TIG1 expression could be observed in the suprabasal layers of epidermis, and there was no expression in the central part of psoriatic lesions. TIG1 expression was less in the basal layer of the marginal part of psoriasis and uninvolved tissues compared with that of the normal skin (P〈0.01). TIG1 expression was also less in the suprabasal layers of the central part of psoriasis compared with that of the normal skin and the uninvolved tissues (P〈0.01). Conclusion differentiation of epidermal keratinocytes and may be associated with the abnormal TIG1 may maintain the normal differentiation of psoriatic keratinocytes.
