Objective To investigate the alteration of cell proliferation and tazarotene-induced gene 3 （TIG3） mRNA expression in cultured normal human keratinocytes after tazarotene treatment. Methods After 24-hour incubation with 0. 1 and 1 μmol/L tazarotene of normal human keratinocytes, the proliferation potency of the cells was detected by MTT colorimetric assay and the expression of TIG3 mRNA was examined by reverse transcription polymerase chain reaction （RT-PCR） and real-time quantitative RT-PCR （TaqMan）. Results After treatment with 0.1 and 1 μmol/L tazarotene,keratinocytes proliferation was inhibited by 5.0% and 11.1%, and TIG3 mRNA expression was induced by 2.1- and 2.8-fold, respectively. Conclusion Tazarotene can in- hibit keratinocytes proliferation and elevate TIG3 mRNA expression.
