目的用聚肌苷酸胞苷酸(polyinosinic polycytidylic acid,PolyI:C)注射C57BIL/6小鼠以研究建立原发性胆汁性肝硬化(PBC)动物模型,探讨CD40配体(CD40L)在外周血T淋巴细胞表面的表达,和在PBC中T淋巴细胞的活化情况。方法20只C57BIM6小鼠随机分对照组和模型组。将polyI:C以5ms/kg的剂量腹腔注射模型组小鼠,对照组注射PBS,每周2次,120d后处死,留取外周血和肝组织,间接免疫荧光法检测血清抗线粒体抗体(AMA),HE染色观察胆管病理变化,生化分析仪测定血清碱性磷酸酶(ALP)含量,流式细胞术分析外周血CD4^+、CD8^+T淋巴细胞所占比例及CD40L在T淋巴细胞表面的表达情况。结果PolyI:C注射120d后模型组小鼠均出现AMA阳性,肝组织小胆管周围有不同程度的炎性细胞浸润,血清ALP明显高于对照组(P〈0.001);对照组胆管及血清ALP均未发生明显变化;模型组小鼠CD40L^+/CD4^+、CD40L^+/CD8^+T淋巴细胞[(4.35±0.34)%,(1.42±0.10)%)显著高于对照组[(0.78±0.10)%,(0.43±0.04)%,P〈0.01];而模型组小鼠CD4^+、CD8^+T淋巴细胞所占比例[(25.83±1.80)%,(24.84±2.70)%]与对照组[(20.51±3.46)%,(17.84±1.53)%]比较,差异无统计学意义(P〉0.05)。结论PolyI:C腹腔注射C57BIM6小鼠120d可引起PBC病变,活化的CD4^+、CD8^+T淋巴细胞在PBC发病中起重要作用。
Objective Polyinosinic polycytidylic acid (PolyI: C ) was used to generate primary biliary cirrhosis animal model in C57BL/6 mice, and CD40 ligand in T lymphocytes was analyzed to show T cell activation. Methods 20 C57BL/6 mice were divided into 2 groups equally, model group and control group. Mice in model group were injected with PolyI:C (5 mg/kg) and PBS twice a week. 120 days after injection they were put to death. Antimitochondrial antibodies (AMA) were detected by indirect immunofluorescence (IIFL), pathology changes were determined by haematoxylin and eosin stain (HE), alkali phosphatase (ALP) was assayed by biochemistry analysator, at last the number of CD4^± T, CD8^± T cells and the CD40 L expression in T cells were analyzed by flow cytometry (FCM). Results After 120 days PolyI:C injection all mouse in model group were found AMA positive and inflammation cell infiltration around bile ducts. However, there were almost no changes in serology and pathology items in contro/ group. In addition, the level of ALP in model group was higher than that in control group (P 〈0. 001 ). The ratios of CD40L^±/CD4^±, CD40 L^±/CD8^± T cells in model group[(4.35 ± 0.34)%, (1.42 ± 0.10)%] were significantly higher than that in control group [(0.78 ± 0.10)%, (0.43 ± 0.04)%, P 〈 0.01 ] . respectively, but the percentage of CD4 and CDs over total T cells in model group [ (25. 83 ± 1.80 )%, ( 24. 84 ± 2. 70) % ] had no significant difference from that in control group [ ( 20. 51 ± 3.46 ) %, ( 17. 84 ± 1.53 )% ,P 〉0.05 ]. Conclusions Primary biliary cirrhosis animal model can be established by injecting PolyI:C in C57BL/6 mouse and activated CD4^+ and CD8^+ T cells are critical in PBC.
