为了研究磷酸八钙骨水泥(octacalcium phosphate,OCP)作为药物载体的体外释药行为,以利福平(rifampicin,RFP)作为模型药物,制备利福平/酸八钙骨水泥药物缓释体系。通过X射线衍射、红外光谱、场发射扫描电镜分析、万能电子试验机及紫外-可见分光光度计等测试方法,研究RFP对OCP的相组成、抗压强度的影响及其释药行为。结果表明:在OCP中添加适量的药物并不会影响最终产物的物相;对于药物含量一定的载体,药物释放时间在5 d后,强度变化趋于稳定;对于药物含量不同的载体,药物含量越高越容易导致载体的孔隙率增大,强度变小;药物含量分别为60 mg/g与40 mg/g的载体的释放特性都符合Higuchi扩散释放模型,但是前者的相关性更好。孙建英’,王德平1,2姚爱华’,周萘’,黄文品1,2
To study drug release behavior of octocalcium phosphate cement(OCP) as a drug carrier,rifampicin(RFP) was chosen as model drug to prepare OCP loaded with RFP(RFP/OCP).The X-ray diffraction,Fourier transform infrared spectroscopy,field emis-sion scanning electron microscopy,electronic universal testing,and ultraviolet-visible spectrometry were performed to characterize variation in the composition,compressive strength,and drug release behavior.The results indicate that a certain amount of RFP has no influence on the hydration properties and final composition;for the same RFP/OCP,the compressive strength becomes steady when RFP/OCP is immersed in simulated body fluid for 5 d.However,for OCP with different amounts of RFP,the increased amount of RFP results in higher porosity and smaller compressive strength.OCP with 60 mg/g and 40 mg/g RFP are both consonant with the Higuchi diffusion release model,but the former is more consonant.
