[目的]探讨疏肝健脾方对抑郁障碍型乳腺癌小鼠脾脏中NKT细胞核与血清IL-13表达的影响。[方法]将4T1炎性乳腺癌细胞(每只5×105个/0.2ml)接种于BALB/c小鼠右侧第二对乳腺脂肪垫内并给予28d慢性不可预知性刺激,建立抑郁障碍型乳腺癌模型,随机分为正常对照组、单纯抑郁组、单纯荷瘤组、抑郁荷瘤组、单纯中药组、单纯化疗组和中药合并化疗组。应用流式细胞术检测各组小鼠脾脏中NKT细胞核的比例变化,运用ELISA方法检测小鼠血清中IL-13表达水平。[结果]BALB/c小鼠接种4T1炎性乳腺癌细胞后NKT细胞核表达比例显著升高(P〈0.01),在合并抑郁障碍后(单纯抑郁组)有升高趋势(P〉0.05);小鼠脾脏中NKT细胞核表达比例与血清IL-13表达水平呈正相关(r=0.617,P〈0.01)。单纯中药组、单纯化疗组、中药合并化疗组小鼠NKT细胞核表达比例及IL-13的表达均明显低于正常对照组(P〈0.01),在中药合并化疗组中两者表达水平最低。[结论]在荷瘤机体,尤其是伴有抑郁障碍的情况下,脾脏内NKT细胞核和IL-13的表达均显著增高。IL-13可能参与了NKT介导的抑郁障碍型乳腺癌免疫重塑过程,而疏肝健脾方可能通过改善脾脏中NKT细胞核IL-13的表达水平发挥治疗作用。
[Purpose] To investigate the effects of shuganjianpi prescription on NKT nucleus and serum IL-13 content in the depression disorder breast cancer mouse spleen. [Methods]The inflammatory 4T1 breast cancer cells(5×105/0.2ml) was inoculated in BALB/c mice on the right side of the second pairs mammary fat pad. The model mouse was given the chronic softly unpredictable stimuli for 28 days to set up the depression breast cancer model. These mice were randomly divided into normal control group,pure depression group,pure tumor group,depression tumor group,pure Chinese medicine group,pure chemotherapy group and Chinese medicine combined with chemotherapy group. The NKT nucleus of the spleen was detected by FCM,the expression of IL-13 in the serum was detected by the ELISA method. [Results] The expression of NKT nucleus in the BALB/c mice which were inoculated inflammatory breast cancer cells 4T1 increased significantly(P〈0.01),and have the same tendency with the depression disorder(P〉0.05),and the expression level of NKT nucleus was positively associated with IL-13(r =0.617,P〈 0.01). The expression level of NKT nucleus and IL-13 in the group of pure Chinese medicine,pure chemotherapy and Chinese medicine combined with chemotherapy was lower than that in the normal control group(all P〈0.01),and in the Chinese medicine combined with chemotherapy group the expression was the lowest. [Conclusions]In the tumor mice,especially with the depression disorder,the expression level of NKT nucleus and IL-13 increased significantly. That indicates the IL-13 worked on the immunity mediated by the NKT nucleus in the depression breast cancer,and the shuganjianpi prescription has the therapeutically effect by changing the expression level of IL-13 in the NKT nucleus of the spleen.
