目的围绕重组乳酸菌的药用优势、发挥药用效应的免疫学机制、影响因素、主要应用及其发展中面临的问题进行综述。方法分析和整理近年来国内外关于口服重组乳酸菌药用研究与应用的相关文献。结果与结论多种乳酸菌以其安全性、遗传可操作性及良好的定植能力成为口服药用蛋白在胃肠道发挥局部作用的潜在运载工具。近10年的动物试验表明,口服重组乳酸菌具有明显的药用价值,目前该项研究已进入人类临床实践阶段。虽然重组乳酸菌口服剂型的研发仍存在技术瓶颈,但其应用前景和潜在的商业价值不可估量。
OBJECTIVE To elaborate the advantages, immunological mechanisms of medicinal effects, influencing factors as well as applications of recombinant lactic acid bacteria (rLAB). In addition, the issues of their development are also discussed. METHODS The literatures about the medicinal research and applications of oral rLAB at home and abroad in recent years were analyzed and organized. RESULTS AND CONCLUSION A variety of LAB have become potential vehicles for oral pharmaceutical proteins on exerting local actions in the gastrointestinal tract (GI tract) because of their safety, genetic manipulation feasibility and colonization. The animal experiments in the past decade have demonstrated the obvious medicinal values of rLAB by oral mucosal administration. The research of oral rLAB has entered the stage of human clinical trials. Although there still exist technical bottlenecks in the development of oral dosage forms of rLAB, the prospects and potential commercial values of the bacteria are inestimable.
