从中国典型培养物保藏中心(CCTCC)引进Marc-145细胞,建立了原始细胞库、主细胞库和工作细胞库,并对工作细胞库的细胞复苏后进行了活力、形态、生长特性、微生物污染、核型、乳酸脱氢酶同工酶和对猪繁殖与呼吸综合征病毒(PRRSV)敏感性等特性进行了鉴定.结果显示,Marc-145细胞为上皮型细胞,生长状态良好,生长曲线呈“S”型,最大增殖浓度为4.71×10^5/mL,倍增时间为26.1h,细菌、真菌、支原体和红细胞吸附检测均为阴性,染色体核型为2n=60,对PRRSV敏感.建立的细胞,为开展猪繁殖与呼吸综合征病毒及其疫苗研究提供了细胞资源.
A strain of Marc- 145 cells was introduced from CCTCC. The cell line was preserved to three cell banks, that is, primary cell bank, main cell bank and working cell bank. Then the cells were thawed from the working cell bank and the studies on cell biological characteristics were carried out, including vitality, morphology, growth dynamic, microbial contamination, karyotype analysis, LDH isozyme analysis and susceptibility to PRRSV. The morphology showed that Marc-145 cells were epithelium- liked cell and grew well. The growth curve was "S" type and the maximum propagation density was 4.71 x 105/mL. The population doubling time (PDT) was 26.1 h. The test results for bacteria, fungi, virus and mycoplasma were all negative. The number of the chromosomes was 60 and diploid cells were dominant. What was important, the Marc- 145 cells showed sensitive to PRRSV. So the established Marc- 145 cell banks were good resources for PRRS virus and relative vaccine research.
