人们很早就发现微量元素硒在生命和人类健康中的作用。硒以硒代半胱氨酸(Sec)形式掺人到硒蛋白中,并以硒蛋白的形式发挥作用。GPx-4是硒蛋白谷胱甘肽氧化还原酶(GPx)家族的一员,其能直接减少脂质过氧化物。近年来研究使我们了解GPx-4在精子形成、精子功能和胚胎发育过程中有着重要作用。而GPx-4基因剔除引起小鼠胚胎致死,GPx-4水平下降与人类不育症和其他许多地方性疾病都相关。本文旨在综述GPx-4的生物学功能和基因多态性方面的研究进展。
The requrement of the trace element selenium for life and its beneficial role in human health has been known for several decades. Selenium in the form of amino acid selenoeysteine (See) incorporated into the selenoprotein. GPx-4 is a member of the classcial glutathione peroxidases(GPx) family,which is a unique antioxidant enzyme that can directly reduce phospholipid hydroperoxide in mammalian cells. Recently, molecular techniques have allowed overexpression of GPx-4 in mammalian cell lines, from which it is becoming clear that GPx-4 has an important role in spermatogensis, sperm function, and embryonic development, and its deficiency is implicated in human infertility ,embryonic lethality of GPx-4 konckout mice and many endemic diseases. In this review,we focus on the biological ruction and polymorphism reserch about various diseases of GPx-4 in mammalian ceils.
