目的研究白藜芦醇(resveratrol)对大鼠急性心肌缺血的保护作用,并进一步阐明其机制。方法建立急性心肌缺血模型,持续观察标准Ⅱ导联心电图30min,6h后进行心肌TIC染色测量心肌梗死面积,测定血清肌酸激酶(CK),乳酸脱氢酶(LDH)活力,确定心肌损伤程度。心肌细胞凋亡的检测采用原位末端标记法(TUNEL),免疫组化法检测凋亡相关基因Fas、Bax及Bcl-2的蛋白表达。结果白藜芦醇能够剂量依赖性地降低大鼠心肌冠脉结扎后ST段抬高,降低CK、LDH活力,缩小心肌梗死面积,降低心肌细胞凋亡指数,抑制Fas、Bax的表达,增强Bcl-2的表达。结论白藜芦醇对冠脉结扎诱发的大鼠急性心肌缺血有保护作用,其机制可能与上调Bcl-2蛋白表达,下调Fas、Bax蛋白表达,抑制心肌细胞凋亡有关。
Objective To study the effects of resveratrol on acute myocardial ischemia induced by ligation of the left anterior descending branch of the coronary artery in rats and its possible mechanisms. Methods Rat acute myocardial ischemia model was induced by ligation of the left anterior descending branch of the coronary artery, then continuously observed limb lead Ⅱ ECG for 30min. Tne infarction sizes were measured 6 hours later after 2,3,5-triphenyhetrazolimn chloride (TIC) staining. Tne activities of serum creatine kinase (CK) and lactate dehydrogenase(LDH) were measured. Tne myocardial cell apoptosis was determined with terminal deoxynucleotidyl transferase-mediated dUTP-fluorescence in nick end method. Bax, Bcl-2 and Fas protein expression were detected by immunohistochemistry and imaged analysis. Results Resveratrol could dose-dependently inhibit the raose of ST segment decrease the activity of CK and LDH, reduce infarction size in acute myocardial ischemic rats, decrease apoptosis index, up-regulate the protein expression of Bcl-2 gene and down-regulate Bax and Fas. Conclusion Resveratrol showed protective effects on acute myocardial ischemia. This effect might be related to modulating the expression of apoptosis related proteins including Bcl-2, Bax and Fas.
