目的 应用miRNA芯片技术研究小分子RNA(miRNA)对犬心房纤颤(AF)的调节作用,发现了AF的新机制,为新药研究提供了新的靶点。方法 将杂种犬随机分为两组,假手术组(n=4);利用高频起搏器对右心耳进行刺激起搏诱发持续性AF组(n=5),取左心房组织常规抽提总RNA,采用含有468种miRNA的芯片检测系统进行miRNA表达谱差异分析。结果 和假手术组相比,AF模型组的miRNA表达谱中差异表达的miRNA共有10个。其中有4个miRNA上调,6个miRNA下调。结论 miRNA参与了持续性AF的调节作用,提示miRNA是抗心律失常药物作用的新靶点。
Objective To investigate the expression pattern of microRNA (miRNA) in atrial fibrillation (AF) using miRNA array technique and search new targets of anti-AF drugs. Methods Nine dogs were randomly divided into sham-operated group( n = 4) and AF model group ( n = 5) in which group a pacemaker was implanted in a subcutaneous pocket and attached to a screw-in epicardial lead in the right atrial appendage. Total RNA was extracted from leftatrium and the differential expression of miRNA was detected by miRNA array. Results Among 468 target miRNAs, ten miRNAs showed differentially expression, four were up-regulated and six down-regulated. Conclusion miRNA is a new target of antiarrhythmic drug since it is involved in the modtdation of continuous AF.
