采用体外培养的方法,研究了2种粒径的石英粉体(KWC-Q3和KWC-Q4)和纳米二氧化硅粉体(Nano-SiO2)对A549细胞存活率、乳酸脱氢酶(LDH)释放、白细胞介素(IL-6)和肿瘤坏死因子(TNF-α)的影响。结果表明Nano-SiO2对A549细胞的毒性作用最大,对KWC-Q3其次,对KWC-Q4毒性最弱;随着暴露矿物粉体浓度的升高,细胞的死亡率明显升高,细胞释放的LDH随之升高,存在剂量效应关系。石英和Nano-SiO2能够引起A549细胞分泌IL-6的增加,但是并未引起TNF-α变化。
The human lung epithelial cell line A549 was exposed to Nano-SiO2and quartz of two kinds of particle size(KWC-Q3 and KWC-Q4) for 24 hours.The effects of them to cell viability,leakage of lactate dehydrogenase(LDH),secretion of interleukin 6(IL-6) and tumor necrosis factor(TNF-α) were studied.This study demonstrated that: comparing with KWC-Q3 and KWC-Q4,Nano-SiO2 in A549 cells performed the highest cytotoxicity.The viability of A549 cells was significantly decreased in a dose-dependent manner after exposure to different concentration of KWC-Q3,KWC-Q4 and Nano-SiO2.The leakage of LDH was significantly increased with the increase of cell mortality.The secretion of IL-6 in quartz-treated cells and Nano-SiO2-treated cells increased compared with the control group.However,there is no detectable difference of TNF-α between treated cells and untreated cells.
