[目的]探讨延边农村地区朝鲜族人群脂联素基因外显子4个位点基因SNPs与不同代谢指标的相关性.[方法]在延边农村地区,筛选出197例代谢综合征(MS)患者作为研究对象,同时选择147例同性别及年龄的非MS人群作为对照组.采用RFLP-PCR技术检测脂联素基因SNP+45,SNP+276,SNP-11377,SNP-11391的基因型,并测定体质量指数(BMI)、腰围(WC)、收缩压(SBP)、舒张压(DBP)、空腹血糖(FBG)、三酰甘油(TG)及高密度脂蛋白胆固醇(HDL-C)等代谢指标水平.[结果]MS组SNP+276位点GG型的WC,BMI,TG,SBP及DBP水平均显著高于GT+TT杂合型(P〈0.05,P〈0.01);MS组SNP-11377位点CC型的WC,BMI,TG,SBP及DBP水平均明显高于CG+GG杂合型(P〈0.05,P〈0.01);MS组SNP-11391位点GG型的WC,TG,SBP,DBP及FBG水平均显著高于GA+AA杂合型(P〈0.05,P〈0.01).MS组SNP+45位点基因型及对照组4个位点基因型不同代谢指标间差异均无统计学意义(P〉0.05).[结论]延边农村地区朝鲜族人群脂联素基因外显子4个位点基因SNPs与不同代谢指标的相关性程度有所不同.
OBJECTIVE To study the correlation between four loci genes of single nucleotide polymorphism(SNPs)in adiponectin gene exon and the different metabolic indexes in Korean-Chinese population of Yanbian countryside.METHODS197 cases of MS and 147 cases of non MS with age and gender matched were selected as MS group and non MS control group.Genotypes of SNP+45,SNP+276,SNP-11377 and SNP-11391 of adiponectin genes were detected by using the RFLP-PCR,and the different metabolic indexes,including BMI,WC,blood pressure,FBG,TG,HDL-C and insulin,were measured.RESULTSIn MS group,the levels of WC,BMI,TG,SBP and DBP were significantly higher in GG genotype than in CG+GG heterozygotic genotype for SNP+276(P〈0.05,P〈0.01),and the levels of WC,BMI,TG,SBP and DBP were significantly higher in CC genotype than in GA+AA heterozygotic genotype for SNP-11377(P〈0.05,P〈0.01),and the levels of WC,TG,SBP,DBP and FBG were significantly higher in GG genotype than in GA+AA heterozygotic genotype for SNP-11391(P〈0.05,P〈0.01),and the different metabolic indexes had no statistical differences between SNP+45loci genotypes in MS group and four loci genes of SNPs in non MS control group.CONCLUSIONThere is different correlation between four loci genes of SNPs in adiponectin gene exon and the different metabolic indexes in Korean-Chinese population of Yanbian countryside.
