目的:研究牙龈蛋白酶对成骨细胞MC3T3-E1的增殖及凋亡的作用,以及BMP-7在此过程中的保护作用.方法:分离提纯牙龈蛋白酶,建立牙龈蛋白酶诱导成骨细胞凋亡模型,使用MTT法、TUNEL/DAPI染色法检测牙龈蛋白酶对成骨细胞增殖及凋亡的影响,并且研究BMP-7对于牙龈蛋白酶造成的细胞凋亡的保护作用.结果:牙龈蛋白酶呈剂量和时间依赖性地抑制成骨细胞增殖,诱导成骨细胞凋亡.在500和1000 ng/ml浓度范围内,BMP-7呈剂量依赖性地减轻牙龈蛋白酶对成骨细胞的增殖抑制及凋亡诱导作用.结论:BMP-7能有效抵抗牙龈蛋白酶诱导产生的成骨细胞增殖抑制及细胞凋亡.
Objective: The aim of this study was to determine the proliferative and apoptotic effects of gingipains on os- teoblasts, and to study the protective effects of BMP-7 on apoptosis induced by gingipains. Methods: Gingipains from por- phyromonas gingivalis W83 was purified and MTT assay and TUNEL/DAPI staining were used to determine the prolifera- tive and apoptotic effects of gingipains and BMP-7 on MC3T3-E1. Results: Gingipains inhibited prolifbration and en- hanced apoptosis ofosteoblasts in a dose and time dependent manner. At certain concentrations (500ng/ml and 1000ng/ml), BMP-7 protected the osteoblasts by promoting proliferation and reducing apoptosis.
