目的:研究双参通冠方对急性心肌梗死模型大鼠血流动力学的影响。方法:SD大鼠分为假手术组、模型组及双参通冠低(2.5g/kg)、中(5g/kg)、高(7.5g/kg)剂量组,结扎冠状动脉左前降支建立大鼠心肌梗死模型,于十二指肠给药后0、15、30、60、90、120、180min,采用PowerLab血流动力学信号采集与分析系统,分别检测大鼠的HR、MAP、LVSP、LVEDP、LVdp/dtmax、LVdp/dtmin c。结果:与假手术组比较,模型组各时间点HR、MAP均升高,LVSP、LVEDP、LVdp/dtmax、LVdp/dtmin降低(P〈0.05)。与模型组比较双参组用药15rain后HR、MAP均降低,LVSP、LV—EDP、LVdp/dtmax、LVdp/dtmin均显著升高(P〈0.05)。双参通冠高剂量组LVSP、LVEDP、LVdp/dtmax、LVdp/dtmax改变与中低剂量组有显著差异(P〈0.05)。结论:双参通冠方能增加急性心梗模型大鼠心脏搏出量、降低心肌耗氧量和血管外周阻力,从而改善心功能。
Objective:To research the effects of Shuang Shen Tong Guan Decoction(SSTGD) on hemodynamics in rats with acute myocardial infarction(AMI). Methods:SD rats were divided into sham operation group, model group, SSTGD low-dose group (2. 5 g/ kg) ,SSTGD middle-dose group (5 g/kg) and SSTGD high-dose group (7. 5 g/kg). The levels of HR, MAP, LVSP, LVEDP, LVdp/ dtmax and LVdp/dtml. were detected by PowerLab at 0, 15,30,60,90,120,180 min after administration. Results:Compared with the sham operated group, the levels of HR, MAP were increased, the levels of LVSP, LVEDP, LVdp/dtmax and LVdp/dtmin were reduced at each detected point in model group, the difference was significant ( P 〈 0. 05 ). Compared with the model group, HR and MAP were decreased in SSTGD low-dose group, SSTGD middle-dose group and SSTGD high-dose group after 15 min;LVSP, LVEDP, LVdp/dtm= and LVdp/dtmin were higher,there was significantly difference(P 〈 0.05 ). Compared with SSTGD low-dose group and SSTGD middle- dose group, the difference of LVSP, LVEDP, LVdp/dtmax and LVdp/dtmin was statistically significant in SSTGD high-dose group ( P 〈 0. 05 ). Conclusiou:SSTGD can improve cardiac function by increasing heart stroke volume, decreasing cardiac oxygen consumption and total peripheral vascular resistance.
