克隆了一个新的人类Lish/CTLH结构域基因,经国际基因命名委员会批准命名为RMND5A.该基因位于染色体2p11.2,长3 239 bp,编码391个氨基酸,其结构域从N端起依次为LisH、CTLH与CRA.RT-PCR分析表明该基因在人类早期胚胎的多个组织中有表达,在心脏、肝和肾中的表达水平较强.对该基因的蛋白进行了表达并制备了多克隆抗体.荧光素酶活性测定分析表明,在COS7细胞中过表达蛋白可以使MAPK信号途径的两个下游转录因子ELK-1和AP-1的转录活性显著下降,表明RMND5A通过MAPK信号途径参与了对细胞生命活动的调控过程.
We cloned a novel human gene which contains Lish/CTLH domain and named RMND5A as approved by HUGO nomenclature committee. This gene is located in chromosome 2p11.2. Its cDNA is of 3 239 bp and coding a polypepeide containing 391 amino acids with a LisH domain, a CTLH domain and a CRA domain from N-to C-terminal. This gene is expressed in most of the tissues of human embryos, especially higher in the heart, liver and kidney. The protein was expressed and used to immunize Rabbits to obtain the polyclonal antibody against RMND5A. Overexpression of RMND5A in COS7 cells may remarkably inhibit the transcription activities of ELK-1 and AP-1 that are downstream effectors of MAPK pathway. In conclusion, RMND5A may be involved in regulation of cell life progress through acting a role in MAPK pathway.
