目的观察宁夏无果枸杞芽提取物(FLE)对衰老小鼠小肠上皮FOXO1、β-catenin表达的影响。方法将60只13月龄自然衰老昆明小鼠分为老年对照组和FLE低剂量组(FLE1组)、FLE中剂量组(FLE2组)、FLE高剂量组(FLE3组),每组15只;另将15只6月龄昆明小鼠做为成年对照组。FLE1组、FLE2组、FLE3组分别用质量/体积百分比为5%、10%、20%FLE灌胃,0.2 m L·10 g^-1·d^-1,老年对照组和成年对照组灌胃等量生理盐水,连续8周。应用PCR技术与免疫蛋白印迹技术(Western-Blot),检测小鼠小肠组织中FOXO1、β-catenin mRNA和蛋白表达。结果与成年对照组比较,老年对照组小鼠小肠组织中FOXO1、β-catenin mRNA与蛋白表达下调(P〈0.05)。与老年对照组比较,FLE1、FLE2 FOXO1、β-catenin mRNA表达无明显增加(P〉0.05);FLE3组的FOXO1、β-catenin mRNA和蛋白表达均上调(P〈0.01)。结论高剂量FLE能够上调小肠组织中FOXO1、β-catenin mRNA和蛋白表达。
Objective To observe the effects of Ningxia fruitless lycium sprout extracts (FLE) on expressions of FOXO1 and β-catenin of small intestine in aging mice. Methods 60 KM mice aged 13 months were randomly divided into aged control group and FLE in low dose group (FLE1), middle dose group (FLE2), high dose group (FLE3) and 15 mice in each group; the other 15 KM mice aged 6 months as adult control group. In group FLE1, group FLE2 and group F LE3 the mice received intragastric administration with a concentration of 5%, 10%, 20% FLE gavage respectively (0.2 ml/10 g · d), aged control group and adult control group were treated with intragastric saline (continue 8 weeks). PCR and Western blot methods were used to detect the expressions of FOXO1 and 13 - catenin in small intestine. Results compared with adult control group, the expressions of FOXO1 and β - catenin mRNA and protein in small intestine were decreased in aged control group (P 〈 0.05 ) ;compared with aged control group, the expressions of FOXO1 and - catenin mRNA and protein in small intestine in FLE3 were up - regulated ( P 〈 0.05 ). Conclusion FLE can increase the expres- sions of FOXO1 and β - catenin in small intestine of mice. its mechanism may be related to FLE improve proliferation and differentiation ability of small intestinal epithelial cell of aging mice through Wnt - β - catenin pathway.
