目的了解引产儿胚胎与成人骨髓CD34+细胞生物学活性,寻找新的造血干细胞来源RUNX1相关基因和Notch信号通路差异。方法RT-PCR检测引产儿胚胎和成人骨髓CD34+细胞RUNX1、Hes-1、SLC9A3R1、Notch1、Notch2、HLA-C、PD-CD1、PKC-βI的mRNA表达水平。结果引产儿胚胎骨髓CD34+细胞RUNX1、Hes-1、SLC9A3R1、Notch1、Notch2、HLA-C、PDCD1和PKC-βI的mRNA表达水平与成人组比较差异均无统计学意义(P〉0.05)。结论引产儿胚胎骨髓CD34+细胞具有与成人骨髓CD34+细胞相似的生物学活性,可以作为实验中正常对照使用,满足实验需要。
Objective To explore the difference of RUNX1 genes and Notch signals in embryo and adult bone marrow derived CD34+ cells.Methods The mRNA expression of RUNX1,Hes-1,SLC9A3R1,Notch1,Notch2,HLA-C,PDCD1 and PKC-βI was detected using reverse transcriptase-polymerase chain reaction(RT-PCR).Results The expression of RUNX1,Hes-1,SLC9A3R1,Notch1,Notch2,HLA-C,PDCD1 and PKC-βI in bone marrow CD34+ cells from human embryo and adult showed no significant difference(P0.05).Conclusion The biological activity of CD34+ cells from embryo bone marrow is similar to those from adult,and can be used as normal controls in experiments.
