高铜对雏鸡脑组织细胞凋亡影响的研究

360羽1日龄艾维茵肉鸡随机分为6组,分别喂以对照日粮(Cu 10.89 mg.kg-1)和高铜日粮(Cu 100mg.kg-1,高铜I组;Cu 200 mg.kg-1,高铜Ⅱ组;Cu 400 mg.kg-1,高铜Ⅲ组;Cu 600 mg.kg-1,高铜Ⅳ组;Cu800 mg.kg-1,高铜V组)6周,以流式细胞术和免疫组化染色检测高铜对脑组织细胞凋亡的影响。流式细胞仪检测,高铜Ⅲ组、高铜Ⅳ组和高铜V组脑组织细胞凋亡比例升高,与对照组比较差异显著或极显著(P〈0.05或P〈0.01)。免疫组化染色检测脑组织Bax和Bcl-2蛋白表达显示,高铜Ⅲ组、高铜Ⅳ组和高铜V组Bax表达阳性细胞比例较对照组极显著升高(P〈0.01),而Bcl-2表达阳性细胞比例较对照组显著或极显著降低(P〈0.05或P〈0.01)。同时,高铜Ⅲ组、高铜Ⅳ组和高铜V组脑组织铜锌超氧化物歧化酶活性显著降低(P〈0.05或P〈0.01)和丙二醛含量显著升高(P〈0.05或P〈0.01)。结果表明,日粮铜含量达到或超过400 mg.kg-1可诱导雏鸡脑组织细胞凋亡。

英文摘要:

The experiment was conducted with the objective of examining the effect of dietary high copper on the cellular apoptosis of brain tissue in the chicken by the methods of flow cytometry (FCM) and immune histochemical staining. 360 one-day-old Avian chickens were randomly divided into six groups,and feed on diets as follows:Control (Cu 10.89 mg· kg^-1) and high copper (Cu 100 mg · kg^-1,high copper group I ;Cu 200 ng·g^-1,high copper group Ⅱ ;Cu 400 ng·g^-1 ,high copper group m ;Cu 600 ng·g^-1 ,high copper group iV ;Cu 800 ng·g^-1 ,high copper groupV ) for six weeks. By FCM,the percentage of apoptotic brain ceils was significantly increased in high copper groups Ⅲ, Ⅳ and Ⅴ compared with that of control group (P〈0.05 or P〈0. 01). By the method of immune histochemical staining,the percentage of positive cells of Bax was significantly increased in high copper group Ⅲ, Ⅳ and Ⅴ (P〈0.01) ,and the percentage of positive cells of Bcl-2 decreased (P〈0.05 or P〈0.01) in high copper group IV and Vin comparison with those of control group. Also, copper-zinc-superoxidase (Cu-Zn-SOD)aetivity was obviously decreased and malondialdehyde (MDA)content markedly increased in high copper groups Ⅲ,Ⅳ and g than that in control group. These results showed that dietary copper in excess of 400 mg · kg^-1 induced the cellular apoptosis of brain tissue in chickens.
